I want to know as how will optimize index does SQL server 2000 EE . How many query per second supported sql server 2000 EE.

Obviously the answer will always depend on the type of query you're running.
But if you're more interested in the theoretical scalability you can check http://www.tpc.org/ for benchmarks.

It will support quite a few, mainly dependent on you storage hardware. There's no good answer to this question as the type of query, complexity, indexing, as well as hardware that changes all the time, move this number around.
tpc.org is a good place to start.

TPC.org is a good place to learn about how people design highly scaleable systems. You have to be willing to wade through 500 page reports, but if you are willing to - you'll see some amazing uses of hardware and start gaining a better understanding of how to physically scale up SQL Server.
